Buffers for SDS PAGE

SDS PAGE basic buffer (x10) - 1 liter

  • Dissolve 30.3g Tris(hydroxymethyl)aminomethane (Tris, Mw=121.14) and 144.1g
    2-(4-hydroxyphenyl)aminoacetic acid     (glycin, Mw=167.16) in 800ml ddH2O.
  • Adjust volume to 1 liter with ddH2O.
  • Store at room temperature.

 

SDS PAGE running buffer - 1 liter

  • Mix 100ml of SDS PAGE basic buffer (x10) with 890ml ddH2O.
  • Add 10ml SDS 10%.
  • Prepare before use.

 

SDS PAGE transfer buffer - 1 liter

  • Mix 100ml of SDS PAGE basic buffer (x10) with 200ml methanol and 700ml ddH2O.
  • Prepare before use.

 

SDS PAGE sample buffer (x2) - 100ml

·         Mix 10ml 1.5M Tris (pH 6.8), 12ml SDS 10%, 30ml glycerol, 15ml
b-mercaptoethanol and 1.8mg bromophenol blue.

·         Adjust volume to 100ml with ddH2O.

·         Divide into 10ml aliquots and store at -20 oC.

·         Store working solution at 4 oC.
